ABSTRACT

ObjectiveTo discuss the characteristics of psychological assistance hotline calls and operators' coping strategies of before and after the COVID-19 outbreak, in order to further improve the assistance ability of the psychological crisis hotline. MethodsA retrospective study was conducted on the demographics characteristics, call problems, coping strategies, and call time trends recorded by Changchun psychological assistance hotline information registration platform before the epidemic in Changchun City (January 20, 2019-April 20, 2019) and during the epidemic period (January 20, 2020-April 20, 2020). ResultsThe differences between gender, age, marital status, location, and occupation type before and during the epidemic were statistically significant (χ2=11.205, 234.240, 152.083, 265.458, 353.385, P<0.01). The number of different help calls had a statistically significant difference before and during the epidemic (χ2=185.088,P<0.01). The difference in the number of operators’ different coping strategies before and during the epidemic was statistically significant (χ2=226.810, P<0.01). Before the epidemic, the main peak of incoming calls was concentrated at 16∶00 to 17∶00, and the secondary peak was concentrated at 22∶00 to 23∶00. During the epidemic, the main peak of incoming calls was also concentrated at 16∶00 to 17∶00, while the secondary peak was concentrated at 10∶00 to 11∶00. ConclusionDuring the COVID-19 epidemic, the number of calls to the psychological assistance hotline was higher than that before the outbreak. The main peak time for calls was the same, and the secondary peak was adjusted from 22∶00 to 23∶00 to 10∶00 to 11∶00. During the epidemic, the number of calls from male, 30 to 39 years old, married, local and staff in Changchun was the most, psychological problems counseling and operator referral strategy were the most before and after the epidemic.

ABSTRACT

Growth hormone (GHRH) and pitutary adenylate cyclase activating peptide (PACAP) are the members of the PACAP/Glucagon superfamily, who are related in both sequence and function.Their stimulation of GH secretion and animal growth is concerned.A series of expression plasmid,pIRES1-GHRH-PACAP (P-G-P),plRESI-GHRH (P-G) and plRESI-PACAP(P-P),were constructed, extracted and purified, then transfected into CHO cell line with Lipofectamine. The expression was examined by RT-PCR,dot-ELISA and Western blotting. The biological activity of expression products was detected in rats.At 8 h after injection of transfection supematant, serum IGF-I concentrations in P-G-P group were significantly higher than that in other groups(P < 0.05).PLGA encapsulating plasmid microspheres were prepared and injected intramuscularly into rabbit legs.Growth behavior and IGF-1 level were measured at day 0,15,30 and 45 after injection.Greater body weights gain and higher serum 1GF- [ levels were observed in three plasmid microsphere injection groups, compared with control group. At day 30,the body weight gain in P-G-P group was greater than saline group (81%,P< 0.01),P-G mierosphere group (15%,P< 0.05) and P-P microsphere group (7%,P> 0.05),serum IGF-I concentration in P-G-P microsphere group showed a 16.68% increase to P-G microsphere (P > 0.05),a 17.14% increase to P-P microsphere(P > 0.05) and a 50.46% increase to control (P < 0.05).These results suggest that co-expression of GHRH and PACAP in one expression plasmid might exert an additive stimulation of GH secretion and growth when delivered into rabbit skeletal muscle with PLGA mierosphere. The results may provide a new approach to regulate animal growth.

ABSTRACT

OBJECTIVETo explore the inhibitory effect of He-Ne laser irradiation on fibroblast growth of hypertrophic scars in culture.

METHODSHe-Ne laser with wavelength of 632.8 nm, power density of 50 mW/cm(2) and doses of 3 J/cm(2), 30 J/cm(2), 90 J/cm(2) and 180 J/cm(2) was used to irradiate human scar fibroblasts in culture 1, 3 and 5 times respectively, and then the cell count and cell cycle analysis were done.

RESULTSRepeated irradiation with He-Ne laser at dose of 180 J/cm(2) three and five times led to an evident decrease in total cell number compared with that of the control group and there was a significant difference (P<0.05). The cell cycle analysis showed after three and five times of irradiation with 180 J/cm(2) He-Ne laser the cell number in S-phase decreased from 51% to 20% and 14% respectively, the cell number in G(0)/G(1) phase increased from 28% to 55% and 60% respectively, and the cell percentage in Sub-G1 phase was 6.7% and 9.8% respectively.

CONCLUSIONSRepeated irradiation with 180 J/cm(2) He-Ne laser can inhibit scar fibroblasts growth in culture. It may be that He-Ne laser irradiation causes cell stagnation in G(0)/G(1) phase and apoptosis.

ABSTRACT

Method We used biomechanical methods in controlling strain of anterior tibial muscles of rabbits .Objective To observe the histological and enzymohistochemical change after sprain.Result Experimental model of muscle sprain could be made with yield load which was about 128% of the body weight extracting anterior tibial muscle of the rabbits. Some of the muscle fibers broke near the junction between muscle and the tendon. Conclusion Fibrosis of endomysium and scar formation at the injury might be an important cause of frequent recurrence of muscle sprain.

ABSTRACT

Objective To establish animal model for hypertrophic scar and study the characters of its morphology and collagen metabolism. Methods A total of 64 round wounds (diameter of 6 mm each) with total skin loss were made on the ventral side of rabbit ear using a trephine. Morphology and collagen metabolism of scar wounds were studied at 14,21,35,70 and 98 days after operation, respectively. Results There were 76% elevated scars developed (45/59 wounds) on the ventral side of rabbit ear at 21 days and 46% elevated scars disappeared (11/24) at 98 days after operation. There were numerous fibroblast proliferation and whorl-arranged collagen fibers at 21 and 35 days. The number of fibroblast decreased, but irregular-arranged fibers still presented in the elevated scars at 70 and 98 days after operation. Hydroxyproline content in elevated scars at 21 days was higher than that in normal skin (P<0.05), and at 35 days was 3 times as that in normal skin and at 98 days was also markedly higher than that in normal skin (P<0.05). Conclusion Excessive deposition of collagen is a characteristic of hypertrophic scar in rabbits. The conversion of normal scarring to hypertrophic scarring in rabbits occurs at 14~21 days after operation. Both development and regression of hypertrophic scar in rabbit are quicker than that in human.
